Azo dye degradation by Phanerochaete chrysosporium in the medium enriched with nitrogen in the presence of primary co-substrate BABT
Andrade,Marcus Vinicius Freire; Silva,Karla Mayara Lima da; Siqueira,João Paulo da Silva; Wanderley,Carlos Ronald Pessoa; Araújo,Rinaldo dos Santos; Marinho,Gloria; Rodrigues,Kelly.
This work sought to evaluate the ability of fungi Phanerochaete chryosporium to degrade congo red azo dye in batch reactor, evaluate the influence of glucose and wheat bran as co-substrates on the removal of the dye in the medium and investigate the influence of ammonium chloride, ammonium nitrate and ammonium sulfate as the inorganic nitrogen source for the process. Wheat bran was not effective satisfactorily for the removal of dye and organic matter had no desired effect for the removal of color and organic matter and showed the lowest values of k2, 0.008 M-1.d-1and 0.0004 M-1.d-1, respectively. Glucose presented the best response with the highest final percentage of dye removal (97%) and rate of dye removal (0.017 M-1.d-1), without adding an external...

Comparative study of a new alkaline L-methioninase production by Aspergillus ustus AUMC 10151 in submerged and solid-state fermentation BABT
Abu-Tahon,Medhat Ahmed; Isaac,George Saad.
Twenty four fungal species were screened for their ability to produce alkaline L-methioninase on methionine-glucose liquid medium. Aspergillus ustus AUMC 10151 displayed the highest yield of enzyme (10.8 U/mg protein), followed by A. ochraceus and Fusarium proliferatum. Upon optimization of the submerged fermentation (SmF)conditions, the maximum enzyme yield (18.23 U/mg protein) was obtained on a medium containing L-methionine (0.5%), sucrose (0.95%), KH2PO4 (0.1%) and 175 rpm. Seven agro-industrial by-products were screened as substrates for L-methioninase production under solid-state fermentation (SSF). Wheat bran resulted 38.1 U/mg protein, followed by rice bran (27.6 U/mg protein) and soya bean meal (26.6 U/mg protein). Maximum alkaline L-methioninase...

Digestive and functional properties of a partially hydrolyzed cassava solid waste with high insoluble fiber concentration Scientia Agricola
Raupp,Dorivaldo da Silva; Rosa,Danielle Amorim; Marques,Silvia Helena de Paula; Banzatto,David Ariovaldo.
Starch factories generate large amounts of cassava solid waste. A small amount is utilized for animal feed but most of it is discharged with deleterious effects to the envirounment. A edible food with a high content of insoluble dietary fiber (60.9%), named "partially hidrolyzed cassava waste" (PHCW), was prepared from industrial cassava solid waste by an enzymatic process. PHCW or wheat bran (WB) were fed to model rats and both promoted digestive function effects, but PHCW produced the greatest effect. The insoluble fiber constituent from PHCW (and not the soluble fiber), promoted the greatest fecal bulking, fecal weight and defecation frequency in rats, as compared to WB. Such results indicate that the partially hydrolyzed cassava waste presents...

Effect of moisture content and inoculum on the growth and conidia production by Beauveria bassiana on wheat bran BABT
Nuñez-Gaona,Oscar; Saucedo-Castañeda,Gerardo; Alatorre-Rosas,Raquel; Loera,Octavio.
The aim of the present work was to study the effect of moisture content and inoculum on the growth and conidia production by Beauveria bassiana on wheat bran (WB).The highest growth rate of B. bassiana 885.2 on WB media was obtained at a w=1.0, with no detected growth at a w < 0.97. Solid-state fermentation (SSF) using WB (66% moisture; a w=1.0) achieved a maximal yield of 1.18x10 10 conidia per gram of dry substrate (gds). This yield decreased one order of magnitude with higher moisture contents or the addition of sugarcane bagasse (SCB) as a texturiser. In SSF using WB (66% humidity),the time to obtain a yield of 1x10(10) conidia/gds, referred to as t10, could be predicted using a model considering common inoculum levels and maximal yields. For...

Enhanced production of Aspergillus tamarii lipase for recovery of fat from tannery fleshings BJM
Dayanandan,A.; Rani,S. Hilda Vimala; Shanmugavel,M.; Gnanamani,A.; Rajakumar,G. Suseela.
The influence of various oil cakes has been investigated for high level production of lipase using Aspergillus tamarii MTCC 5152. By solid state fermentation in wheat bran containing 2.5% w/w gingili oil cake at 70% v/w moisture content the fungus produced a maximal yield of lipase (758 ± 3.61 u/g) after 5 days of incubation using 2% v/w inoculum containing 10(6) spores/mL. Wheat bran and gingili oil cake with supplementation of gingili oil (1.0% w/w), glucose (0.5% w/w) and peptone (0.5% w/w) gives an increased enzyme production of 793 ± 6.56 u/g. The enzyme shows maximum activity at pH 7.0, temperature 50 °C and was stable between the pH 5.0-8.0 and temperature up to 60 °C. Crude lipase (3%) applied to tannery fleshing shows 92% fat solubility. The...

Novel process for the simultaneous extraction and degumming of banana fibers under solid-state cultivation BJM
Jacob,Nicemol; Prema,Parukuttyamma.
Various process parameters for the production of polygalacturonase by Streptomyces lydicus under solid-state fermentation were optimized. The optimum particle size of wheat bran for polygalacturonase production was in the range of 500-1000 µm. Initial moisture content of 70% was found to be the optimum for enzyme production. The most suitable inoculum size was 1.25 x 10(5) CFU/mL and the optimum incubation temperature was 30ºC. Addition of carbon sources resulted in 37% increase in enzyme yield (425 U/g), whereas no significant enhancement was obtained on nitrogen supplementation. Maximum enzyme yield was recorded at 72 h. When compared to the initial production medium (108.5 U/g), the enzyme yield was 3.9 fold after optimization. Solid-state fermentation...

Scale up of production in a bioreactor of a halotolerant protease from moderately halophilic Bacillus sp. isolated from soil BABT
Prasad,Roopa; Abraham,Theruvath Koshy; Nair,Ananthakrishnan Jayakumaran.
Studies were conducted on the production of protease by moderately halophilic Bacillus sp. on agro-industrial waste materials. The bacterium could efficiently use many agro wastes as substrates but wheat bran supported maximum enzyme production. To ascertain the performance of the process in shake flasks and lab scale bioreactor, experiments were conducted to analyse protease activity utilizing wheat bran as cost effective substrate. The studies unveiled that pH 7.0, temperature 30°C and static conditions were optimal for enzyme production in flask level fermentation. In scale-up fermentation, at optimal pH and temperature, agitation rate of 50 rpm was best for protease production. The enzymatic nature was studied in 10% SDS gels with BSA (2.5 mg/mL) as...

Sources of carbohydrates in the ingestive behavior of feedlot steers R. Bras. Zootec.
Silva,Viviane Santos da; Alves Filho,Dari Celestino; Freitas,Leandro da Silva; Brondani,Ivan Luiz; Restle,João; Azevedo Junior,Ricardo Lima de; Teixeira,Odilene de Souza; Borchate,Daniele.
In this article we research the influence of different sources of carbohydrates (corn, soybean hulls or wheat bran) upon the digestive behavior of 24 confined castrated steers with an initial average age and weight of 20 months and 330 kg born from the cross between Charolais and Nellore. The diet was composed of 40% sorghum silage and 60% concentrate. The time spent on total ruminating (an average of 454.6 min/day) was not influenced by the source of carbohydrate. The animals from the wheat bran treatment spent less time idle (718 min) in relation to those on the corn and soybean hulls treatments, which did not differ between themselves (an average of 792 min/day). The steers from the wheat bran treatment remained less time feeding (184 min/day) compared...

Sperathe effects of solid-state fermentation in the functional properties of defatted rice bran and wheat bran BABT
Silveira,Cristina Moreira da; Badiale-Furlong,Eliana.
Functional properties of fermented bran produced by Aspergillus oryzae and Rhizopus sp. in a solid-state fermentation system were determined, with an aim to evaluate their application in food formulation. The defatted rice bran and wheat bran were inoculated with the spores of the cultures and incubated at 30º C for 72 h. Samples were withdrawn at 0, 24, 48 and 72 h. Protein content, protein solubility, in-vitro digestibility, gelation and water holding capacity were determined in bran with or without fermentation. Rhizopus sp. increased significantly the protein content (69.0 and 56.0%, respectively, for defatted rice bran and wheat bran); protein solubility (28.5 and 36.2) and water holding capacity (11.4% for wheat bran). When A. oryzae was used all...

Wheat bran- but not oat bran-enriched diets increase the mucosal height of the cecum and colon of newly weaned and aged rats BJMBR
Cavaglieri-Felippe,C.R.; Polacow,M.L.O.; Campos,M.R.; Vecchia,M.G.; Curi,R..
The effect of diets enriched with oat or wheat bran (prepared by the addition of 300 g of each fiber to 1000 g of the regular diet), given for 8 weeks, on the mucosal height of the colon and cecum was investigated. Newly weaned (21 days old) and aged (12 months old) male Wistar rats were used in this study. As compared to controls, diets enriched with wheat bran provoked a significant increase in the mucosal height, whereas oat bran did not cause any effect. In newly weaned rats (21 days old), wheat bran increased the mucosal height (µm) in the cecum by 20% (mean ± SEM for 8 rats; 169.1 ± 5.2 and 202.9 ± 8.0 for control and wheat bran, respectively) and in the colon (218.8 ± 7.2 and 264.5 ± 18.8 for control and wheat bran, respectively). A similar effect...

Xylanase production by a thermo-tolerant Bacillus species under solid-state and submerged fermentation BABT
Gupta,Uma; Kar,Rita.
Effects of xylose on xylanase production by a thermophilic Bacillus sp showed diverse patterns on corn cob (CC) and wheat bran (WB) as sole carbon sources in solid- state fermentation (SSF) and submerged fermentation (SmF). Supplementation of these media with either mineral salt solution (MSS) or yeast extract peptone (YEP) also exerted variable effects. While under SSF, xylose stimulated xylanase synthesis by 44.01%, on wheat bran supplemented with MSS, it decreased the enzyme activity by 12.89% with YEP supplementation. In SmF, however the enzyme synthesis was stimulated by xylose on supplementation with both MSS and YEP by 41.38% and 27.47%, respectively. On corn cob under SSF, xylose repression was significant both with MSS (26.92%) and YEP (23.90%)...
